LC-MS TOF Salient Characteristics
Liquid Chromatography–Mass Spectrometry (LC/MS)
1. Must be capable of both small molecule compounds and large molecule biomarker/proteomic determinations. Have the ability to perform both qualitative and quantitative analysis.
2. If bench top model, must include workbench.
3. Nitrogen generator must be provided with the system.
4. Computer hardware and software must be included. Chemical analysis software must include MS and MS/MS capabilities. Data analysis tool must be able to identify differences in type and amount of metabolites present and be able to data mine from large complex data files using mass and/or mass spectra. Must be able to analyze with a database of known compounds.
5. Hands on training course with future onsite consulting must be available
6. System must be capable of future hardware additions/upgrades i.e. ESI and APCI or gas phase.
7. Software must have an automated tune procedure for optimization of ion optics and full mass axis calibration. It should have data dependent capabilities of dynamic selections of fragmentation energy, preferred charge state, and exclude/include mass list.
LC
1. The LC system must be high pressure injection system for fastest LC separation.
2. The LC System must have an automated multi-sample loading system.
3. The LC System must have High efficiency orthogonal ion spray source to increase target ions with fewer solvent particles.
4. The LC System must have Binary pump to include degasser, solvent selection valves, cabinet, bottles, and all required connections.
MS
1. The MS System must be capable of performing Time of Flight TOF determination. Quad or Triple TOF acceptable.
2. The MS System must have Low thermal expansion flight tube to reduce mass accuracy error from surrounding room temperature changes. Mass error should not be more than 1 ppm per 1 degree C.
3. The MS System must have Built in calibrant delivery system for automatic introduction of reference mass into analytical sample for real-time mass correction.
4. The MS System must be able to complete a cycle switching for positive to negative.
5. The MS System must feature automated optimization of ion optics and full TOF mass axis calibration. Ion beam compression that enables simultaneous maximum sensitivity and resolution without having to change modes.
6. TOF detector must offer a minimum five decades in-spectrum dynamic range without data manipulation.
7. The MS System must have a spectral acquisition rate in both MS and MS/MS modes of 50 spectra/second.
