PURCHASE DESCRIPTION
1. Statement of Need and Purpose:
The Infections of the Nervous System Section (INSS) of the National Institute of Neurological Disorders and Stroke (NINDS) requires multiple reagents, reagent kits, and consumables to be used with an existing Xenium Analyzer for in situ analysis of genes that provides subcellular localization and quantitation of RNA transcripts within fresh frozen or fixed tissue and the analysis of spatial transcriptomics in the HIV infected brain.
Analysis of RNA transcripts could also help identify biomarkers, which will help the INSS gain a better understanding of HIV infection and other neurological disorders.
2. Background Information and Objective:
The mission of the INSS lab is to study the Neuropathogenesis of HIV infection, Amyotrophic lateral sclerosis (ALS), Multiple Sclerosis, and other neurological disorders.
Currently, there is no available treatment to prevent or hinder HAND in chronically infected HIV patients, which represents a gap of knowledge within the field of HIV-1 research. The INSS lab is trying to identify the cell type responsible for the HIV Central Nervous System (CNS) reservoir and assessing viral transcription dynamics within these reservoir cells as well as exploring transcriptomic changes in the reservoir cells and their neighbors. The INSS lab found that viral RNAs can be detected in the CSF and serum of HIV-infected individuals using a series of digital droplet PCR assays that have been designed to assess viral transcription dynamics. Furthermore, the INSS found that the level of viral RNAs correlates with neurocognitive impairment as measured by a global deficit score and cognitive domain scores for verbal fluency, memory, executive functioning, and learning in patients under long-term antiretroviral therapy. Additional viral RNAs and proteins produced from low levels of basal viral transcription can be released from the cell and impact neighboring cells by activating the innate immune response. These findings suggest that further research is needed to understand the mechanisms that contribute to neurocognitive dysfunction in infected individuals and arrest these cascades to achieve a functional cure for HIV infection. The purchase of the reagents and consumables for Xenium Analyzer will allow for spatial transcriptomic analysis of HIV brain autopsy tissue to address unmet need in the field. Analysis of RNA transcripts could help identify biomarkers and gain a better understanding of HIV infection in the brain. Such profiling and identification of potential biomarkers will be performed for neurological diseases such as HIV, amyotrophic lateral sclerosis (ALS), Multiple Sclerosis, Alzheimer’s Disease, Parkinson’s Disease, gliomas and is important for the NINDS mission, to reduce the burden of neurological diseases.
