Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose: This acquisition is necessary for the multiome sequencing analysis of brain tissues for RNA analysis experiments which support neurodegenerative disease research within the Center for Alzheimer’s and Related Dementias (CARD).
2. Background Information and Objective:
The National Institute on Aging (NIA), Center of Alzheimer’s and Related Dementias works to understand the pathological basis of neurodegenerative diseases mainly Alzheimer’s Disease. The objective of this acquisition is to provide a multiome sequencing instrument for analysis of RNA changes associated with Alzheimer’s Disease and other neurodegenerative disease disorders. A multiome sequencing instrument is a powerful molecular analysis device that allows for the simultaneous detection of analytes, including single-nucleotide variants (SNVs), copy number variants (CNVs), and proteins.
3. Generic Name of Product: Multiome sequencing instrument
4. Purchase Description:
Description:
Cat no: Quantity
Tapestri instrument MB012-0020 1.00
Tapestri single-cell DNA + protein training kit MB03-0050 1.00
Tapestri single-cell DNA training kit MB03-0049 1.00
5. Salient characteristics: All deliverables required under this contract shall be packaged, marked and shipped in accordance with Government specifications. At a minimum, all deliverables shall be marked with the contract number and Contractor name. The Contractor shall guarantee that all required materials shall be delivered in immediate usable and acceptable condition.
Instrument must include these specifications:
The platform must utilize droplet microfluidic technology to rapidly encapsulate, process, and profile 5,000 to 10,000 individual cells for single-cell DNA analysis.
The platform must utilize a two-step workflow, including (1) cell encapsulation and protease incubation to allow access to DNA within individual droplets and (2) barcode addition and target amplification.
Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
The platform must allow for target amplification without use of whole genomeamplification methods (e.g. multiple displacement amplification).
The platform must have the capability to do both cell encapsulation and barcode/reagent addition on the same cartridge.
The workflow must take no more than 4 hours of hands-on time from cell suspension to sequencing-ready libraries.
The platform must allow for robust detection of single-nucleotide variants (SNVs), small insertion/deletions (indels), and copy number variants (CNVs) across regions targeted by gene specific primers.
The platform must be able to simultaneously detect SNVs, CNVs and cell surface proteins from the same cell.
The platform must be capable of detecting subclones with ≥2 co-occurring variants at 0.1% and de novo subclones at 1% of the population.
The instrument must fit compactly on a standard laboratory benchtop with a small footprint of approximately 1 sq ft and weighing approximately 14 lb/6.35 kg.
The instrument must have a simple and intuitive user-friendly touchscreen interface to run the cell encapsulation and cell barcoding/reagent addition steps of the workflow.
Panels must have panel coverage uniformity of >80% and reads on-target of >80%.
