GENERAL INFORMATION

Title of Acquisition and Project:

Acquisition: Whole genome sequencing services using 90 human iPSC samples Project: NCATS study of effect of passaging and CEPT cocktail on ES and iPS cell genomic stability.

Statement of Need and Purpose:

The SCTL requires whole genome sequencing services to be performed by the vendor using 90 SCTL human iPSC samples. The data generated from this services will be utilized by SCTL scientists to study the effects of cell culture conditions on genomic instability.

Background Information and Objective:

The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.

The National Center for Advancing Translational Sciences (NCATS) is a part of the National Institutes of Health (NIH), with the mission to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions. The Stem Cell Translation Laboratory (SCTL) is a state-of-the-art research facility within NCATS’ Division of Pre-Clinical Innovation dedicated to addressing the scientific and technological challenges in the iPSC field.

The SCTL uses an integrated technological approach to better understand pluripotent stem cell biology and thereby leverage drug discovery and regenerative medicine.

The SCTL aims to develop novel cell culture conditions that reduce genomic instability and increase the safety of iPSC technologies. The whole genome sequencing services provided by Genewiz are an integral part of the suite of analysis methods that will be carried out to construct a high-resolution “map” of the genomic and epigenomic landscape that controls human pluripotency. The integration of all these analyses will allow us to generate foundational cell culture methods that will be shared with the scientific community.

Period of Performance:

The period of performance is 45 days from receipt of the samples from NCATS.

SCOPE OF WORK

General Requirements:

Independently and not as an agent of the Government, the contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Specific Requirements listed below.

Specific Requirements:

Whole Genome Sequencing: This service performs genome-wide sequencing (WGS). WGS is designed to study mutation frequency, genetic variants or variants associated with disease and/or treatment conditions.

The contractor shall:

1. Provide protocols to NCATS SCTL for preparation of the samples.

2. Perform whole genome sequencing services on 90 human samples. This includes implementing customized steps to purify genomic DNA, and the whole genome sequencing.

The samples must be sequenced at ~100X coverage (~270Gb of data) using the same platform used for previous sequencing for NCATS.

3. Provide the deliverable listed below.

GOVERNMENT RESPONSIBILITIES

NCATS SCTL will provide the contrctor with 90 human cell samples. NCATS anticipates, but does not guarantee, providing the samples to the contractor in September of 2020 in one (1) shipment. Using a suitable common carrier, all cells will be shipped overnight using dry ice.

SCTL staff will prepare samples according to protocols provided by the contractor.

DELIVERY OR DELIVERABLES

The contractor will submit the following deliverables to NCATS SCTL in electronic format:

• Raw data (FASTQ) output files.

• Summaries and documentation

REPORTING REQUIREMENTS

The contractor must alert the Government to any issues that arise which should be brought to the attention of the Government.

OTHER CONSIDERATIONS

Data Rights:

Data generated as a result of all services will be the properties of NIH, SCTL, NCATS

Publications and Publicity:

The contractor will not publish any result of the work.

Confidentiality of Information:

The Contractor will keep the results of the contract confidential.
