2.0 BACKGROUND
The National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS), Genomic
Technology Section (GTS) provides genome-scale, high throughput sequencing services to basic and clinical researchers within the institute. The Genome Analysis Core Facility (GACF), like others at the National Institutes of Health (NIH) must implement emerging cutting-edge technology to maintain the leading edge on genomics and epigenetic research areas.
To support the research of NIAMS Intramural Research Program (IRP), GTS needs to acquire reagents compatible with Chromium Controller Single-Cell Platform, an instrument acquired in
2020 (75N92020P00151) to perform single cell RNA-Seq. NIAMS Investigators task the core facility with single-cell analysis services using the platform’s molecular barcoding and analysis suite that deliver many mRNA transcripts per run. The Platform uses special reagents to produce epigenetic information of thousands of cells of up to eight different cell populations. These components will continue to support several research projects carried out to better understand the genetic determinants of autoimmune, auto inflammatory, and musculoskeletal diseases and gain insights into the pathogenesis of genetic and immunologic diseases at a single-cell level. The platform and its reagents have been configured to be compatible with 10X Genomics Chromium
Controller to support a broad range of applications critical to the research.
