Text version
Requirements of Digital Droplet PCR machine for US National Arboretum, FNPRU:
• Must use droplets as a partitioning method (as opposed to physical chip partitioning), which provides high sensitivity and precision to measure target DNA molecules.
• Must be compatible with EvaGreen DNA binding chemistry that we are currently using for established protocols in the lab, enabling current probes and protocols to be directly transferred between assay types.
• Additional dye compatibility and instrument capability is also required to allow simultaneous detection and differentiation of two (and preferably more) probes, including TaqMan assays.
• Generates 20,000 uniform nanoliter-sized droplets, which are analyzed independently, resulting in high precision measurements especially for rare transcripts.
• Delivers an absolute count of the number of target DNA sequences for applications that include copy number variation, mutation detection, and gene expression analysis, especially to detect rare transcripts.
• Ability to recover DNA from un-analyzed droplets for parallel external cloning and sequencing to complement quantitation results.
