Line Item 0001: Small Footprint Benchtop Flow Cytometer Hardware and Software

SPECIFICATIONS For the Procurement of an Ozone Calibrator for an Inter-Comparison

I. BACKGROUND INFORMATION

The Material Measurement Laboratory’s Biomaterials Group portfolio includes efforts to develop measurements and standards to support advanced cell and gene therapy products and regenerative medicine applications. The Group’s cell characterization project involves development of methods and measurement assurance tools to support the quantification of mammalian cell number and assessment of cell viability and health metrics. A compact, benchtop flow cytometer is needed to complement other existing measurement capabilities in the Group. Flow cytometry is a routinely applied cell quantification technique that uses light scattering and fluorescence emission along with fluorescent dye-based assay kits to obtain quantitative data at the single-cell level. These assays may be commercially available from multiple sources or developed in-house at NIST. Due to laboratory space limitations, a compact, user-friendly flow cytometer for cell quantification and cell health analysis is needed.

II. SPECIFICATIONS

mailto:robert.singman@nist.gov

The flow cytometer system shall consist of an instrument (hardware) containing the computational capabilities and software necessary to run the instrument and analyze the data.

Hardware specifications:

1. The hardware shall consist of a flow cytometer capable of analyzing single cells with software and computational abilities included within the instrument.

2. The size of the flow cytometer shall be no larger than 12 in (height) x 12 in (width) x 12 in (depth).

3. The optics shall consist of a single laser excitation source at 532 nm and 3 detection channels consisting of two fluorescent channels and a forward scattering channel.

5. The instrument shall be able to analyze cells over a range of sizes, including cells ranging from 2 microns to 40 microns in diameter.

6. The instrument shall be capable of quantifying cell samples at dilutions in an approximate range of 104 cells/mL to 5x105 cells/mL.

7. The run time per sample should be no more than 2.5 minutes per sample.

8. The computer system contains at least 150 GB storage.

9. The instrument shall have a screen to display results.

Software specifications:

10. The software shall include experimental and analysis templates for each of the available assay kits from the instrument manufacturer.

11. The software shall have flexibility to accommodate running and analyzing experiments using assay kits from other manufacturers, customized assay kits, and NIST-developed cell characterization assays for viability and apoptosis of cells.

12. The data analysis software shall be available for download to at least one alternate PC system for off-line analysis.

13. The instrument should have capability for USB download of data including graphs and .csv output files for analysis in other graphical programs.
