About this file

This document includes a purchase description and federal contract opportunity for a Multiphoton Mesoscope Imaging System. The National Institute of Mental Health requires one Multiphoton Mesoscope Imaging System from Thorlabs, or an equivalent product, to enable functional imaging within a 5mm field of view over multiple cortical areas in mice, rats, and marmosets. The purchase description provides details on the system's capabilities such as scanning multiple non-contiguous regions within the field of view, a 20 degree rotation and fine XYZ motion, and a remote focusing mirror for fast axial control over 1mm. Offerors should be able to deliver the system within 90 days of order receipt and warrant it for a minimum of one year. The solicitation is issued as a request for quote under simplified acquisition procedures with a not-to-exceed threshold.

PURCHASE DESCRIPTION

Multiphoton Mesoscope Imaging System

1. Statement of Need and Purpose:

The section on Critical Brain Dynamics intends to purchase a Multiphoton Mesoscope Imaging System which is an innovative venture for the advancement of 2-photon and 3-photon imaging research for the National Institute of Mental Health (NIMH). This advanced technology will allow one to record simultaneously from multiple cortical areas with cellular spatial resolution at high, video rate temporal resolution. With this technology, the Plenz and Histed Laboratories will utilize the latest developments of genetically encoded indicators (for imaging) and opsin (for stimulation). To do so, the Meoscope will be equipped with specific lasers that cover a wide range in wavelengths from blue to deep red (~400 – 1300 nm) at high and low repetition rates.

NIMH expect this technology to have a major scientific impact in the near future. NIMH is confident that this technology can bridge two major technological approaches solidly established at the NIMH:

1. Studying human brain dynamics by analyzing inter-areal connectivity in cortex at low spatial resolution, e.g. using fMRI and MEG.

2. Studying animal brain dynamic by analyzing intra-areal connectivity in cortex at high spatial, i.e. cellular resolution using multi-photon imaging.

The Mesoscope will allow laboratory to study inter-areal connectivity at cellular spatial resolution in transgenic animals including mice, rats and marmosets.

2. Background Information and Objective:

The Mesoscope embodies groundbreaking technology pioneered by the Svoboda Lab at Janelia farm, HHMI in conjunction with Thorlabs Inc., VA. The technology allows one to record simultaneously from multiple cortical areas, at different cortical depths, at cellular spatial resolution and high, video rate temporal resolution. Given the many cortical areas participating in even simple sensory and behavioral tasks, NIMH anticipate that this technology will become essential to study adequately the flow of inter-areal information processing in the brain of animals.

Several NIMH research labs identified the usefulness of the Mesoscope for their respective experimental approach:

1. Understanding the simultaneous involvement of multiple cortical areas in the encoding and decoding of sensory stimuli and behavioral outcome in transgenic rodents (Histed and Plenz Laboratories).

2. Tracking of whole cortex presynaptic inputs to single, identified neurons using retrograde viral tracing techniques in transgenic rodents (Unit on Functional Neural Circuits).

3. Interareal functional connectivity at cellular resolution in transgenic marmosets (Transgenic Core Facility).

Technology development:

All-optical interrogation of the brain is the future of neuroscience. This encompasses select, optical stimulation of neurons or groups of neurons and optical imaging of identified neuronal population. The Mesoscope is currently not optimized to allow for select optical stimulation over multiple cortical areas.
