About this file

This document outlines specifications for two quantitative polymerase chain reaction (qPCR) systems. The Agricultural Research Service of the Department of Agriculture seeks to procure one two-target qPCR system equivalent to Bio-Rad's CFX Duet Real-Time PCR System, with the ability to detect two DNA targets per well in a multiplex assay. Specifications include excitation/emission wavelengths of 450-580nm, a temperature range of 4-100°C, and the ability to detect one copy of target sequence. Additionally, one five-target qPCR system is required equivalent to Bio-Rad's CFX Opus 96 Real-Time PCR System, with standalone capabilities to detect up to five DNA targets per well in a multiplex assay. Specifications include excitation/emission wavelengths of 450-684nm and 515-730nm respectively, and the same temperature and detection requirements. Both systems must include appropriate software and reagents.

Attachment 1 – Specifications

USDA ARS SJVASC-CDPG requires two qPCR-Real Time systems (RT-qPCR), one of which can detect two DNA targets in a multiplex qPCR assay and the other capable of detecting up to five DNA targets in a multiplex qPCR assay.

Performance Specifications and Salient Characteristics (Brand Name or Equal):

Two Target RT-qPCR System, equal to BioRad CFX Duet Real-Time PCR System

This instrument must be capable of processing 96 reactions, with multiplex processing of two target genes/well simultaneously. The Real-Time System needs to use Fluorescence Resonance Energy Transfer (FRET) optics to detect target DNA amplicons which increases accuracy and the reliability of data.

SPECIFICATIONS of Two Target qPCR Real-Time System:

Reliable 96-well qPCR system with quick setup, easy installation, and factory-calibrated optics Multiplex analysis – up 2 DNA targets/well (plus FRET) simultaneously without the need for reference/normalizing dye Excitation/emission wavelength range covering 450-580nm Optical detection – excitation (3 filtered LEDs), detection (3 filtered photodiodes) Average ramp rate of at least 3.3⁰C/sec Thermocycler lid temperature range of 30-110⁰C Temperature range of 4-100⁰C Sensitivity: Capable of detecting one copy of target sequence in human genomic DNA Accuracy of +/- 0.2⁰C or better at 90⁰C Ability to process reaction volumes of 1-50µL Dynamic range of ten orders of magnitude Software (CFX Maestro v2.3 or equivalent) for Windows and Mac, with license Uniform optical system free of regular calibration requirements Audit and e-signature compatibility One year warranty iScript Reverse Transcription Supermix, SsoAdvanced Universal SYBR Green Supermix, and iTaq

Universal SYBR Green Supermix included

Five Target RT-qPCR System, equal to Bio-Rad CFX Opus 96 Real-Time PCR System

This instrument must have standalone features, and be capable of processing 96 reactions, with multiplex processing of up to five DNA targets (plus FRET) simultaneously.

Specifications of 5 Target qPCR Real-Time System:

Standalone capabilities (functional without hooking up to a computer) to be used for experiments in gene expression (without requiring ROX), end-point analysis for sequence

12905B23Q0157 Parlier, CA detection, and protein thermal shift assays Touchscreen interface Simultaneous multiplex processing of up to 5 target DNA sequences FRET Detection (Fluorescence Resonance Energy Transfer) of target DNA Excitation/Detection: 6 filtered LEDs/ 6 filtered photodiodes Excitation wavelength range covering 450-684nm Emission wavelength range covering 515-730nm Average ramp rate of at least 3.3⁰C/sec Thermocycler lid temperature 30-110⁰C Temperature range 4-100⁰C Accuracy of +/- 0.2⁰C or better at 90⁰C Sensitivity: capable of detecting one copy of target sequence in human genomic DNA Dynamic range of 10 orders of magnitude Ability to process reaction volumes of 1-50µL Uniform optical system free of regular calibration requirements Software (CFX Maestro v2.3) for Windows and Mac, with license included Audit and e-signature compatibility iScript Reverse Transcription Supermix (or equivalent), SsoAdvanced Universal SYBR Green

Supermix, and iTaq Universal SYBR Green Supermix One year warranty
