Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: Purchase of bench top Flow Cytometer laboratory system with a high sensitivity detector is required for analysis of aspect ratio, size, and fluorescent intensity of cells and isolated mitochondria in a 96 well format, at a high flow rate (up to 20,000 events/s). The unique design required for this instrument should include use of a CCD camera with lasers, as opposed to the less sensitive photomultiplier tubes. The increased sensitivity is needed in particular for analysis of isolated mitochondria in which signals are dim. This CCD camera design enables high sensitivity detection which is a requirement for this instrument. The instrument is needed to determine the effect of Parkinson’s disease-causing genes on mitochondrial function, consistent with the mission of the Inherited Movement Disorders Unit in the Neurogenetics Branch of the NIH.

2. Background Information and Objective: Will need an instrument that can analyze aspect ratio, size, and fluorescent intensity of cells and isolated mitochondria in a 96 well format, at a high flow rate (up to 20,000 events/s). It will enable assessment of mitophagy, mitochondrial reactive oxygen species production, ATP production, among other critical mitochondrial functions. This will aid the research being done by the Inherited Movement Disorders Unit (IMDU) in the Neurogenetics Branch of the NINDS.

They have found that specific genes causing Parkinson’s disease influence the degradation and function of mitochondria. These processes can be monitored in an unbiased way by flow cytometry. Some assays require analysis of isolated mitochondria.

These place special demands on the flow cytometer as the signal is too dim to be detected by conventional flow cytometers that use photo-multiplier tubes. By contrast, the systems with CCD based camera detection have the necessary sensitivity to make these measurements reliably. Additionally, the CCD-camera based system can provide information on parameters like particle size and aspect ratio which will greatly facilitate IMDUs line of research.
