Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose:
The National Institute of Neurological Disorders and Stroke (NINDS) is establishing a new laboratory that will use cutting-edge physiological, imaging, and behavioral methods to investigate the relationship between human neurological disorders and brain function at the molecular, synaptic, cellular, and circuit levels. The new laboratory requires equipment high throughput whole slice/wide-field imaging system with both brightfield and fluorescence imaging capabilities.
2. Background Information and Objective:
The NINDS will soon establish the Laboratory of Circuits, Synapses, and Molecular Signaling (LCSMS), which will be led by the new Scientific Director. A key component to the research studies within LCSMS will be high throughput wide-field imaging of large regions and serial slices for whole-brain quantification, and/or multi-region quantification in both brightfield and fluorescence that is endogenous and/or virally transduced. The whole slide scanner system will be used to capture high-quality images of fixed tissue or dispersed cells for ongoing studies investigating learning and memory, peripheral pain disorders, and neurodegenerative disease.
3. Generic Name of Product:
Whole Slide Scanner System
4. Salient characteristics
The NINDS LCSMS requires a whole slide scanner system to be used to capture high-quality images of fixed tissue or dispersed cells for ongoing studies investigating learning and memory, peripheral pain disorders, and neurodegenerative disease. The system should consist of a wide-field whole slide scanner system with capability for dry, oil, and silicone oil immersion objectives with capability for optical sectioning (Z-acquisition), and an automatic oil dispenser. The system must also contain an sCMOS camera, an integrated CMOS camera, and complete image acquisition software. The specific requirements are detailed below grouped specifically as: (I) The scanner system requirements, (II) The sCMOS camera brand name or equal to Hamamatsu ORCA Fusion requirements, (III) The CMOS color camera requirements, (IV) The image acquisition and image analysis software requirements
I. The whole slide scanner system must satisfy these requirements:
A. An enclosed chamber to house both the microscope and slide holding tray once loaded to avoid ambient light exposure to samples prior to, during, and after imaging.
B. A maximum system capacity of 6 slides (1 slide tray) that can be scaled up in-field
C. A slide loading tray that can accommodate 1X3 in, 2X3 in, 3X4 in, and 4X5 in glass slides/plates
D. A loading mechanism that grabs the slide loading tray and not the glass slides E. Potential to scan the full slide area (55X26mm) with advanced tray concept technology F. A microscope that accommodates both dry, oil, and silicone oil immersion objectives G. The microscope shall contain the following objectives: 2X achromatic, 4X X-APO (NA of 0.16 at a minimum; pixel resolution of 1.369um/pixel), 10X X-APO (NA of 0.4 at a minimum; pixel resolution 0.548um/pixel; working distance 3.1mm) 20X X-APO (NA of 0.8 at a minimum; pixel resolution 0.274 um/pixel), 40X (NA of 0.95 at a minimum;
pixel resolution of 0.137um/pixel; working distance 0.18mm), and 40X silicone oil immersion (NA 1.25 at a minimum; pixel resolution of .137um/pixel; working distance 0.30mm)
H. An automatic oil dispenser for automatic application of application of silicone oil for 40X silicone immersion objective
I. An 8 position FL turret with motorized mirror turret, motorized filter wheel J. A fluorescence white light LED light scoure, Excelitas X-cite XYLIS to allow for addition of filter cubes allowing maximum flexibility if channel choice K. A sedat penta-set filter set cube allowing for imaging of
(DAPI/FITC/TRITC/CY5/CY7)
L. Additional filter cube for CFP, YFP, and mCherry
II. sCMOS Camera, brand name or equal to the Hamamatsu Orca Fusion Camera, must satisfy the following requirements:
A. Diagonal 21mm 2304x2304 6.5um sCMOS, 31.6 fps (USB3.0) 89 fps (CoaXP), Cmount, incl. USB3.0 card, USB3.1A-microB CBL, power supply, one trigger cable, HCImage Live SW
III. The CMOS Color camera must satisfy the following requirements:
A. Integrated 2/3 CMOS camera with pixel size of 3.45um X 3.45um (high sensitivity and high resolution)
IV. The image acquisition and analysis software and workstation must meet these requirements:
A. The software must control all motorized elements B. Automatic sample detection, barcode detection and reading, automatic focus mapping (with manual option included), automatic scanning, automatic stitching, pause and resume scanning, Z-stack imaging, acquire data into multiple image formats (vsi and TIFF), conversion between multiple image formats (vsi, jpeg, TIFF), synchronized multi-image display, stepless zoom, slide browse while scanning, slide annotations, screen capture, oil dispenser fill indicator, silicone oil objective maintenance indicator
C. Software must allow for custom scripting through drag-and-drop interface and for on-the-fly or post-acquisition batch analysis.
D. 2D/3D deconvolution processing of data. All PSFs must be factory-measured specifically for each objective and the deconvolution process must utilize graphical processing unit (GPU) processing for speed.
E. Multiplex scan mode to realign slides for reimaging and align multiple fluorescent channels with a reference channel.
F. Suite of analysis tools, including binary processing and segmentation, ROI analysis, object count and object measurement
G. Seamless integration of brightfield and fluorescence imaging mode with various 3rd party hardware devices (including barcode readers, server integration, and slide-storage extension tower) in user-friendly interface
