About this file

This purchase description outlines requirements for a 3D MINFLUX microscope to be acquired by the National Institute of Neurological Disorders and Stroke. The microscope must have two MINFLUX lasers at 561nm and 642nm wavelengths, provide molecular localization precision of a few nanometers in three dimensions, and include an active sample stabilization module for nanometer precision positioning in xyz axes. Additional key requirements include an inverted microscope body, scanning stage, quad scanning optics, deformable mirror unit, high-power lasers, beam shaping modules, objectives, epifluorescence capabilities, and detection channels. The contractor must deliver the system within 22 weeks of award to the specified National Institutes of Health location and complete installation within 70 days of receipt. A one year warranty is required.

PURCHASE DESCRIPTION

1. Statement of Need and Purpose:

The National Institute of Neurological Disorders and Stroke (NINDS), Synaptic Transmission Section (STS) performs fluorescence protein-based experiments for 1) imaging the nanostructures at the cell membrane and 2) tracking the dynamics of a variety of proteins during vesicle exocytosis and endocytosis in the cells. These experiments need to resolve sub-10 nm range of proteins and membrane, which require ‘Super-Resolution Microscopy’ with Minimal Photon Fluxes (MINFLUX).

2. Background Information and Objective:

Synaptic Transmission Section combines physiological and cell biological approaches to understand the dynamics of cell membrane and a variety of proteins during vesicle exocytosis and endocytosis in living cells at nanometer scale, e.g. dense core vesicle docking, curvature generation and endocytosis. Our mission is first to characterize these processes with super-resolution imaging technique, and then determine the underlying regulating molecular mechanisms involved in neurodevelopmental and neurodegenerative disorders.

3. Generic Name of Product: 3D MINFLUX microscope
