Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

BRAND NAME OR EQUAL

Stellaris 8 Confocal Microscope with Tunable Laser

1. Statement of Need and Purpose:

The Surgical Neurology Branch (SNB), NINDS needs a confocal fluorescence microscope which is essential for conducting our mission critical work which includes the following general lines of inquiry: assessment of the blood-brain barrier in adult and pediatric CNS malignancies;

interrogation of the dynamic role of primary cilia in GBM tumorigenesis and treatment resistance; evaluation of the molecular and cell biological underpinnings of epilepsy; and investigation of subcellular regulation of tumorigenesis in pituitary adenomas. Technical requirements including the ability to simultaneously image using the widest range and flexibility of fluorophores with rapid yet gentle image acquisition to preserve specimen integrity.

2. Background Information and Objective:

Confocal microscopy is a critical tool for probing biological systems. While there are several confocal microscopes within NINDS, these are housed primarily in Building 35 (NINDS Core) or individual labs which creates barriers to access, particularly in experiments involving live cells.

The more capable instruments also have limited availability and together, these limitations mean that in practical terms, the PI signatories have inadequate access to this critical tool.

As this system would extend the range of capabilities of the NINDS microscopy offerings, we propose a shared model in which the SNB PIs would have a block of 50% of the instrument time (during regular hours) with the instrument otherwise open to NINDS investigators for full use.

3. Generic Name of Product:

Confocal Fluorescence Microscope

4. Purchase Description:

Equipment brand name or equal to the Stellaris 8 Confocal Microscope shall be provided to simultaneously image using the widest range and flexibility of fluorophores with rapid yet gentle image acquisition to preserve specimen integrity.

5. Salient characteristics The vendor shall provide equipment brand name or equal to the Stellaris 8 microscope that has the following characteristics:

Minimum Salient characteristics: The vendor shall provide equipment brand name or equal to Leica Microsystems STELLARIS 8 with the following material elements:

1. Prism-based spectral scan head with Tandem Scanner that includes a tunable-speed conventional galvo-scanner and an 8Khz resonant scanner that allows for fast switching between each. The linear scanning needs to acheive speeds up to 2600 Hz or 5200 Hz bidirectionally, scan formats up to 8192x8192 pixels, continuous zoom and continuous scan field rotation, 10 frames per second x 5 simultaneous channels for 50 images per second at 512x512 resolution. The 8Khz resonant scanner must achieve 140 images per second (5 simultaneous channels at 28 fps) and allow for continuous scan field rotation and zoom.

2. Must include the STELLARIS White Light Laser (WLL) Extreme, which is able to excite from 440nm to 790nm with 8 different wavelengths simultaneously, each excitation line being tunable in 1nm increments; laser pulsing allows for lifetime imaging with all detectors (described in #4 below). Also must include a 405nm laser for DAPI excitation and photo-activation with ROIs.

3. the Dynamic Acousto-Optical-Beam-Spitter (AOBS), which instantaneously adapts to each of 8 laser lines simultaneously, has over 95% transmission across the full visible spectrum, and avoids the need for dichroic filters

4. Five (5) simultaneous fully multi-spectral detection channels, including 3 solid state HyD S detectors with sensitivity from 410nm up to 850nm, and 2 GaAsP detectors for imaging wavelengths from 410nm to 750nm, along with transmission light detection

5. the TauSense Family of modalities, including TauContrast, TauSeparation, and TauGating, which use fluorescence lifetime information to generate images, add multiplexing options beyond the standard spectral options, identify auto-fluorescence, and eliminate coverglass reflection

6. LAS X Software, including Leica LAS X Lightning Expert for maximizing signal over noise in order to resolve separate signals that are as close together as 120nm laterally and 200nm axially -- this provides near real-time confocal super resolution capabilities and allows for offline reconstruction -- and LAS X 3D Visualization software module for 3D reconstruction. Additional features include the Navigator basic package for defining regions and positions for high resolution scans with stitching, overview images of any shape, and spiral scanning.

7. Premium CUDA workstation with Intel Xeon Gold 6244 processor, 96 GB of working memory, Nvidia Quadro RTX 5000 graphics card, 256 GB SATA SSD system drive, 2 TB M.2 SSD hard drive for temporary data storage, and 6 TB HDD drive for data storage

10. Four (4) microscope objectives, including

-HC PL APO CS2 10X/0.40 NA

-HC PL APO 20X/0.75 NA CS2, 0.62 mm working distance -HC PL APO 40X/1.30 NA Oil with 240 microns FWD

-HC PL APO 63X/1.4 NA Oil Objective

11. DMi8 fully automated inverted microscope and components, including an optically encoded XY-motorized scanning stage

12. Must include a Super Z Galvo Stage for high speed Z series acquisition and real-tim

XZ scanning

13. Must include an Okolab Stage-Top incubation system with temperature, CO2, and humidity control that fits in the Leica Super Z Scanning Stage.

These features meet the government’s primary minimum requirement of having the ability to multiplex probes for confocal fluorescence imaging of olfactory epithelium and other nervous system tissues using tunable lasers, simultaneous spectral detection channels, fluorescence lifetime information, and a conventional tunable-speed scanner at 10x, 20x, 40x, and 63X magnification and 8,192 x 8,192 resolution. Additionally, the LAS X Software with Lightning Expert meets the government’s minimum requirements of resolving signals as close together as 120nm (offline), 3D reconstruction, and semi-automated scanning of a whole slide with Z-stacking.
