PURCHASE DESCRIPTION

1. Statement of Need and Purpose:

The Neuromuscular and Neurogenetic Disorders of Childhood Section at NINDS studies cellular and animal models of neuromuscular diseases and is in need of a confocal fluorescence imaging system with the specifications described in this document.

2. Background Information and Objective:

At the Neuromuscular and Neurogenetic Disorders of Childhood Section, within the Neurogenetic Branch of the National Institute of Neurological Disorders and Stroke, the lab is developing cellular and animal models of early onset neuromuscular diseases, including diseases in which the primary defect is located at the neuromuscular junction, the myomatrix, or within the myofiber.

Our two main focuses are to 1) study disease pathomechanisms and 2) conduct preclinical testing of gene- and transcript-directed therapeutic strategies. Our ultimate goal is to translate innovative therapeutic approaches from the bench to the bedside, and to provide disease-modifying therapies to patients affected with these rare conditions.

Confocal microscopy allows imaging of multiple fluorescently labeled epitopes within cells and tissues. This experimental approach is instrumental to our studies, to visualize structures in situ and assess localization of specific proteins of interest. Our current instrument, the Leica TCS SP5, is a confocal system with optoacoustic splitter and hybrid detectors, used for variety of applications including some limited live imaging. We are looking into upgrades to this system to include super resolution imaging, 3D imaging options, and lifetime/live imaging sensors. These upgrades are necessary to address some of the challenges we are facing with our current system (limited sensitivity and number of probes, lack of 3D reconstruction, limited live imaging, etc.).

3. Generic Name of Product:

Confocal laser scanning microscopy system
