FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose:

The laboratory of Circuits, Synapses, and Molecular Signaling (LCSMS) at National

Institute of Neurological Disorders and Stroke (NINDS) uses cutting-edge imaging, physiological, and behavioral methods to investigate the relationship between human neurological disorders and brain function at the molecular, synaptic, cellular, and circuit levels. The laboratory requires an episcope imaging system with both brightfield and fluorescence imaging capabilities.

2. Background Information and Objective:

The new established LCSMS at NINDS studies fundamental issues of synaptic transmission, neural signal transduction, and functional mapping of neural circuits relevant to human neurological disorders, such as Alzheimer’s and other neurodegenerative disorders. A key component to the research studies within LCSMS is imaging of fixed brain slices or cultured neurons in both brightfield and fluorescence that is endogenous and/or virally transduced. The episcope will be used to capture high-quality images of fixed tissue or dispersed cells for ongoing studies investigating learning and memory, peripheral pain disorders, and neurodegenerative disease.

3. Generic Name of Product:

Epifluorescence Microscope

4. Salient characteristics:

This requirement is issued on a brand name or equal basis to the Nikon T I2-E Inverted Microscope.

The NINDS LCSMS requires an Epifluorescence Microscope to be used to capture high-quality images of fixed tissue or dispersed cells for ongoing studies investigating learning and memory, peripheral pain disorders, and neurodegenerative disease. It must meet these requirements:

Camera Integration

1. The system must be compatible with an existing Photometrics 95B camera (PRIME 95B

CMOS 1200X1200 41FPS 16BIT with dual back-thinned sCMOS detectors) in the lab.

The system must come with an “integration module/kit” to ensure hardware compatibility and integration with the existing photometrics 95B camera.

2. The software must allow configuration and use of the existing Photometrics 95B camera.

Microscope Body

3. An inverted, motorized microscope platform shall include the following:

a. A required Z cam-link motor

b. Motorized DIC sextuple nosepiece

c. Motorized 6 position epi fluorescent turret

d. Motorized port selector

4. Intelligent 1.5X tube lens

5. The microscope must include the following objectives.

a. CFI Plan Apo Lambda 4X NA 0.2; WD 20 mm or an objective of equivalent or superior resolution

b. CFI Plan Apo Lambda 10X NA 0.45; WD 4 mm or an objective of equivalent or superior resolution

c. CFI Plan Apo Lambda 20X NA 0.75; WD 1 mm or an objective of equivalent or superior resolution

d. CFI Plan Apo Lambda 40XC NA 0.95; 0.17-0.25 mm or an objective of equivalent or superior resolution

e. CFI Plan Apo Lambda 60X oil NA 1.4, WD 0.13 mm or an objective of equivalent or superior resolution

6. The objectives must include Nikon’s proprietary “Lamda” or “Nano Crystal Coat” or similar for increased light transmission and low refractive index.

7. The lenses should be compatible and exchangeable with other Nikon based systems in the lab – specifically, the Ti2 E – CSU-W1 SoRA super resolution confocal

8. An inverted, motorized microscope platform

a. A required Z cam-link motor

b. Motorized DIC sextuple nosepiece

c. Motorized 6 position epi fluorescent turret

d. Motorized port selector

9. The field of view (FOV) of the inverted microscope to the two side camera ports (through left- and right-side) must be > 25 mm each. Fly-eye lenses must be built into the light path to provide uniform illumination from edge to edge.

10. The camera ports shall be compatible with large FOV (25mm) color camera upgrade for future applications

11. The microscope must be upgradable to laser-based applications, such as point scanning confocal, spinning disk confocal, TIRF, or additional camera-based detection

Fluorescense/Light Imaging Components

12. LED light source for transmitted diascopic light including a motorized shutter

13. LED light sources for episcopic light including a motorized shutter

14. The LED epifluorescent light source must be a white light source and include an internal shutter, which can be controlled in software.

15. Fluorescent cubes must be accommodate the large field of view of the inverted microscope (> 25mm)

16. The turret must include the following fluorescent cubes/spaces for imaging:

a. DAPI filter set or 375nm equivalent

b. GFP filter set or 480nm equivalent

c. DS-Red filter set or 560nm equivalent

d. Texas Red or filter set mCherry-fluorophore equivalent

e. CY5 filter set 620nm equivalent

f. Empty space for brightfield imaging

Workstation and Software

17. A PC is to be included with this system that is at least 32 GB RAM and 2x2TB HDD storage. A 27” LCD monitor must be included as well.

18. Software must control all motorized and camera components (see above, must integrate with an existing Photometrics 95b Camera) for image acquisition.

19. The software brand name equivalent to NIS-Elements or equal shall include a comprehensive list of analysis tools including ROI analysis, multichannel acquisition, time lapse, measurement, counting, intensity measurement, denoise.ai (artif icial intelligence module) and full documentation for scale bars and annotation of the images.

20. The software must include Extended Depth of Focus module or equal for making Z volume images with cameras.
