Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

Imaging Cytometer System, Dr. Maric Dragan, NINDS

1. Statement of Need and Purpose: The National Institute of Neurological Disorders and

Stroke (NINDS) Flow and Imaging Cytometry Core Facility (FICCF) provides routine use of conventional flow cytometry, imaging cytometry and fluorescence-activated cell sorting (FACS) technology in assaying a multitude of biological properties at the molecular, subcellular and cellular levels, as specified by NINDS and other NIH intramural investigators. To this end, the NINDS FICCF owns flow cytometry and imaging equipment which requires periodic upgrades or replacements to ensure full operational performance to optimally support continuoulsly evolving research requirements performed at the Facility. FICCF currently needs to replace the existing end-of-life imaging flow cytometer system in order to continue to provide optimal services to NINDS and other Institutes at NIH, which collaborate with NINDS.

2. Background Information and Objective: NINDS FICCF has a need to provide routine use of imaging cytometry technology in order to support over 250 basic and clinical research projects in 45 NINDS and other intramural programs. The system that is currently in use is a highly complex system combining high-throughput microscopic imaging of cells through a flow cell. The purpose of this acquisition is to replace the existing ImageStreamX imaging flow cytometer that has been deemed end of life and is no longer supported by the manufacturer.

The equipment to replace the current system needs to be state-of-the-art system, which will enable the Facility to expand the capabilities of this technology to support novel imaging cytometry applications and maintain backwards compatibility with existing imaging cytometry techniques that have been developed and are routinely performed at the Facility.

4. Purchase Description: Imaging Cytometer System is needed with the specifications listed in the salient characteristics section below.

5. Salient characteristics: NINDS FICCF has a requirement for a benchtop, multispectral, imaging cytometer system designed for the acquisition of 12 channels of cellular imagery. By collecting large numbers of digital images per sample and providing a numerical representation of image-based features, this equipment allows for the

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs combination of the per cell information content provided by standard microscopy with the statistical significance afforded by large sample sizes common to standard flow cytometry. With the requested system, fluorescence intensity measurements will be acquired with the following specifications needed in the system in order to handle the volume of samples the Core Facility receives for analysis and to ensure that optimal results are obtained:

• full UV-NIR light spectrum excitation capability that includes 7 lasers with following laser line options 375 nm, 405 nm, 450 nm 488 nm, 561 nm, 592 nm, 642 nm, 730 nm and/or 785 nm

• full UV-NIR light spectrum emission detection capability with commonly used fluorophores including DAPI, Alexa Fluor 405, Alexa Fluor 430, Alexa Fluor 488, Alexa Fluor 546, Alexa Fluor 594, Alexa Fluor 647, IRDye 680LT, IRDye 800CW, and PerCP

• high-resolution, high-QE camera with large field of view imaging capability

• up to 12 imaging detection channels including fluorescence and brightfield

• confocal, epifluorescence and extended depth of focus (EDF) imaging capability

• support multiple high NA microscope objectives including 10X, 20X, 40X, 60X

• autoloader compatible with standard 96-well plates

• integrated image cytometry analysis software with machine learning capabilities

• data analysis import/export compatibility with flow cytometry standard (FCS) and image cytometry experiment (ICE) formats

7. Delivery Date: Within 30 days of purchase. Delivered to Building 10, Room 5S241

8. Other Requirements: The system will come standard with a one-year, on-site warranty.

This includes all travel, parts and labor. The warranty shall not include periodic maintenance and service of the equipment. The Contractor shall provide an Original Equipment Manufacturer (OEM) certified service technician to perform setup, installation, and an on-site training session for staff members.
