PURCHASE DESCRIPTION

1. Statement of Need and Purpose: The purpose of this acquisition is to procure a High-

Resolution FluoRespirometry system that allows high-resolution data regarding oxygen consumption of isolated mitochondria or cells with simultaneous measurement of membrane potential, H2O2 production, or calcium for the Inherited Movement Disorders Unit (IMDU) of the Neurogenetics Branch (NGB) of the NIH.

2. Background Information and Objective: This instrument is needed for research projects in the Inherited Movement Disorders Unit (IMDU) of the Neurogenetics Branch (NGB) of the NIH. This Instrument allows high-resolution data regarding oxygen consumption of isolated mitochondria or cells with simultaneous measurement of membrane potential, H2O2 and has been used as a standard which has improved understanding of mitochondrial respiratory control and the pathophysiology of mitochondrial diseases production, or calcium. This is needed for assessment of mitochondria in our cellular and mouse models of Parkinson’s disease. 4. Specify model, type, catalog, and/or part number(s): Catalog #:10002-03, O2k-

FluoRespirometer: for High-Resolution FluoRespirometry incl. the TIP2k-Module and O2k-sV-Module

5. Salient characteristics:

The Contractor shall provide the NINDS with a High-Resolution Fluorescent Respirometry system brand name or equal to the Oroboros O2K-FluoRespirometer and shall satisfy the specifications below.

I. Hardware and Software

• Must allow measurement of two samples simultaneously

• The instrument shall allow for simultaneous measurement of oxygen consumption and fluorescence

• Must have stainless steel housing that permits injection of drugs during stable measurement of oxygen consumption and fluorescent measurements with constant stirring of the sample

• Peltier thermostat 4 to 47 °C; ±0.002 °C

• Barometric pressure transducer

• 2 Duran glass chambers

• 3 polarographic oxygen sensors (POS), with spare

• 2 Amperometric oxygen channels

• 2 ISE sockets for ion sensitive electrodes

• 2 Sockets for optical signals or NO

• 2 PVDF stirrers, electromagnetic control

• 2 PEEK stoppers

• 2 Smart Fluo-Sensors, green LED emission, photodiode and filters, excitation LED 525 nm, precalibrated intensity

• 3 Filter Sets (Mg green/Ca Green, Amplex Red, Safranin) consisting of LED filters and photodiode filters

• Software with free follow-up upgrades within the warranty period

II. Accessories and Consumables

• 1 ISS-Integrated Suction System, to remove medium from the O2k- Chamber

• microsyringes; fixed needles with rounded tips (6 x 10 mm3 and 3 spare plungers, 6 x 25 mm3, 1 x 50 mm3, 1 x 100 mm3, 1 x 500 mm3), with syringe labels, 2 x syringe racks with syringe collars, 2 x tube racks

• Mitochondrial Respiration Medium. For a final volume of 250 mL (typically €1.4/run)

• Cell permeable succinate (25 mg) and malonate (10 mg)

III. Add-on for two-channel operation with the Respirometer with automatic control by

Software of programmable titration regimes and feedback control

• 4 Microinjection syringes, 200 mm3, 180 mm needle length, 0.12 mm inner Ø;

fixed needles with rounded tips

• 200 Needle Spacers, silicone stops with mounting tool

• Needle Safety Support with cable guide

• 10 Filter Papers

IV. Small chamber volume to reduce the sample amount needed for OXPHOS analysis and cellular/mitochondrial respiratory function

• 2 Duran glass chambers, with smaller operation volume (V) of 0.5 cm3 (0.5 mL)

• 2 PVDF stirrers for small volume chambers, electromagnetic control

• 2 PEEK stoppers for small volume chambers

• 2 Black POM chamber holders with O-ring and V-ring

• 2 Black POM POS-holders

V. Must include 2-year parts and labor warranty

VI. Remote support provided by manufacturer

VII. End User Training
