Statement of Work
Background
The current set of technologies used in developing arthropod diagnostic methods at the USDA APHIS Mission Lab includes the use of sequencing and fragment analysis of DNAs. Regions targeted with Next Generation Sequencing methods produced via PCR (polymerase chain reaction) provide massive amounts of data that can be mined for regions that can discriminate among closely related arthropods. These methods are used downstream for pest captures at US ports and by domestic programs in states like California and Florida. Instrumentation that verifies the size and quality of the products produced via PCR is an essential part of QC. Fragment analyzers are used to “proof” the quality of the products being developed by USDA CPHST laboratories. Each fragment (allele) is accurately measured and can vary to other alleles by only a few bases. Subtle differences in DNA, characteristic among closely related species, can then be detected and used in making regulatory decisions or creating policy by the USDA APHIS.
Required Specifications of Product
1. The intent of this statement of work is to purchase one Bio-analyzer/Fragment Analyzer.
2. The methods developed for projects at the USDA APHIS Mission Lab require the evaluation for quality of DNAs used in the development of diagnostic methods for the detection and identification of agricultural pests. This includes the evaluation of nucleic acids for quality as well as for determining fragment length, such as in genotyping of microsatellites.
3. The instrument should perform analyses on DNAs that represent library construction, RNAs, cDNAs, and microRNAs that occur in organismal genomes used as references for methods development. The sensitivity of the instrument must allow for <50pg/uL. The instrument shall allow for 48 samples (48 capillaries) to be analyzed at once.
4. Include low volume samples (20uL) with flexibility in sampling number made possible by interchanging number and types of capillaries. This will reduce waste and provide optimal results. The resolution for distinguishing among DNA fragments, microsatellites, should resolve at three base pairs being detected and having the ability to export results through a MS Windows 7 compatible software interface and have a footprint that fits within 18 x 28 inch space is recommended.
5. Any installation and training concerning the user interface and mechanical operations is recommended in the purchase of the instrument. Additional items needed for the operation and should include consumables for initial tests and analyses for the operation of the instrument, all necessary literature to allow for user operation. A Service Extended Warranty (1 year) coverage is required. This warranty should be good for one calendar year from the date of purchase. This should include technical support via phone and email for hardware and software troubleshooting. Response for any repairs covered through warranty should be reasonably quick, within 48 hours to 7 working days.
