About this file

This is a Single Source Justification (SSJ) form from the U.S. Fish and Wildlife Service for a sole source acquisition of an Illumina MiSeq i100 Plus sequencing system. The document justifies purchasing the equipment from Illumina Inc. for the Whitney Genetics Lab's metabarcoding Early Detection and Monitoring program with the Northeast Fishery Center. The justification is based on the MiSeq i100 Plus being proprietary equipment only available from Illumina, and the lab's existing protocols and established work using Illumina sequencing kits requiring continued use of their specific technology.

The procurement is valued under the Simplified Acquisition Threshold ($250,000) and will be conducted under FAR Part 13 procedures. The document indicates the lab has negotiated a 20% discount and has an established maintenance contract with Illumina. The acquisition is critical to the USFWS Fisheries Aquatic Invasive Species program, allowing cross-lab validation and the ability to update sequencing capabilities for growing research needs. The procurement notice allows interested parties three calendar days to demonstrate competitive advantages, with a response deadline of 7/11/2025 at 5:00pm Central Time.

1. Description of Need

3. Other Capable Sources

2. Market Research Conducted

Pursuant to FAR subpart 13.106-1(b)(1)(i) and (ii), for purchases not exceeding the Simplified Acquisition Threshold (SAT), Contracting Officers may solicit from only one source if the circumstances deem only one source is reasonably available (e.g., urgency, exclusive licensing agreements, brand name, or industrial mobilization).

Instructions: The information in the text boxes are provided as sample language or to indicate a needed fill-in. Sample language should be removed from the final document. All text boxes require information and shall not be left blank.

Text3: The MiSeq i100 Plus is proprietary equipment and is only available by Illumina Inc.
Text2: As this program has been established using Illumina kits due to their capability and reliability, WGL must continue to use the same sequencing kits as NEFC for cross-lab validation and accuracy. This sequencer will replace the discontinued legacy model and will have the ability to be updated for growing needs.
Text1: In Region 3, Whitney Genetics Lab (WGL) is collaborating on a metabarcoding Early Detection and Monitoring (EDM) program with the USFWS Northeast Fishery Center (NEFC, Region 5). The program is being developed to aid FWCOs as a more efficient part of their monitoring efforts to aid in the early detection of AIS. This work is critical to the mission of the USFWS FAC program to Manage Aquatic Invasive Species (USFWS FAC Strategic Goal #3). It is essential that WGL follow established protocols using Illumina sequencing kits to maintain data compatibility across two USFWS Regions, therefore, this is a sole brand request. To increase sequencing efficiency and capacity, reduce risk of contamination, and ensure our technology is cutting edge, we will be purchasing a MiSeq i100 Plus to replace the discontinued MiSeq legacy model.
Text4: If there were a company that could meet the specifications outlined above, WGL would still need to perform additional laboratory validations, compare results against current performance, and would then need to perform additional cross-laboratory validations. This additional testing would significantly delay results of this mission critical project. Currently there are no other sequencers capable of the performance required by the protocols outlined above.
Text6: We have worked directly with our sales representative at Illumina for the best cost of this equipment, including a 20% discount for having a MiSeq legacy model.

Text5: As outlined above, the metabarcoding protocols developed by NEFC and WGL utilize the Illumina MiSeq legacy model, which is proprietary to Illumina Inc. WGL currently has an Illumina MiSeq and NextSeq and has an additional service contract with Illumina for maintenance of this equipment. All maintenance has been performed by Illumina service representatives on a regular schedule and their technicians have quickly and efficiently repaired any issues that have risen over the years.
