STATEMENT OF WORK (SOW)
GENERAL INFORMATION
Title of Project:
LATS Native Kinase Full Panel Profiling
Project Purpose or Objectives:
In this experiment, we treat cancer cell line models of interest with several candidates LATS1/2 inhibitors that we are currently developing, and we will then profile the activity of these drug candidates against the entire human kinome, directly within the treated cells. This will enable us to validate the specificity and potency of our molecules in relevant, physiological settings and will also enable us to identify possible, undesired off-target effects. The kinase platform will profile the activity of given inhibitors in any cell or tissue lysate. This experiment will support a drug development project focused at developing novel inhibitors for the “hippo pathway” kinases LATS1 and LATS1.
LATS1/2 are 1st in class targets, for which no drugs are currently available. Inhibition of LATS1/2 is expected to be beneficial in multiple settings/indications, spanning from regenerative medicine to cancer immune-therapy.
Background Information:
The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.
The National Center for Advancing Translational Sciences (NCATS) is a part of the National Institutes of Health (NIH), which mission it is to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions.
Period of Performance:
July 2019 through July 2020. Work to be completed within 30 days after vendor receives batch samples.

SCOPE OF WORK
General Requirements:
Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below:
Specific Requirements:
Profile fifty-seven compounds of NCATS choice at a concentration we determine in a lysate assay. Profiling will be performed with the ATP Probe directly in the sample provided. The Contractor shall provide full length kinase kinome screening.
Level of Effort:
GOVERNMENT RESPONSIBILITIES
Government will provide cell pellet that is 10 ml and contains 7500-15000 million cells.
DELIVERABLES AND REPORTING REQUIREMENTS
All data should be summarized and included in a final electronic report, deliverable to NCATS via email within a month after the receipt of research materials.
Data Rights:
All data generated, obtained, and analyzed under this contract is the sole intellectual property of NCATS. Vendor will retain no intellectual property.
