(16) Description: The National Institutes of Health (NIH), a part of the U.S.

Department of Health and Human Services, is the nation’s medical research agency — making important discoveries that improve health and save lives. The mission of the (NIAMS) is to support research into the causes, treatment, and prevention of arthritis and musculoskeletal and skin diseases; the training of basic and clinical scientists to carry out this research; and the dissemination of information on research progress in these diseases. One of the main focuses of the Somerman Laboratory, or the Laboratory of Oral Connective Tissue Biology (LOCTB), is the identification of factors crucial toward periodontal development and maintenance. The periodontal complex includes mineralized tissues (alveolar bone and cementum) and non-mineralized tissues (e.g. the periodontal ligament, sandwiched between the alveolar bone and cementum). Destruction of the periodontal complex causes adverse sequelae, including early tooth loss.

The laboratory has identified genes/proteins involved in periodontal development, e.g. progressive ankylosis protein (ANK) and ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1), both important regulators of pyrophosphate. Currently, the lab is working on identifying the roles of these genes in root formation and cementogenesis using mouse models and studies of individuals with ENPP1 mutations. Ablation of either ANK or ENPP1 causes a striking cementum phenotype in the mouse, which is also demonstrated in humans with ENPP1 mutations. The results have shown the importance of pyrophosphate during mineralization in the oral cavity. Ultimately, the lab aims to use the knowledge gained from these studies toward clinical diagnoses and therapies, and thus, the lab is delving into the mechanisms in which ANK and ENPP1 regulate cementogenesis.
