we submit the following responses for the following microscopes –

Objective Preferences:
Overall Notes: Unless otherwise noted, objectives for compound microscopes (i.e., excluding stereo microscopes) must be fully compatible with all compound microscopes in the facility, including the existing Ni-E upright microscope. This allows sharing of high-value objectives rather than purchasing all objectives of interest for all microscopes. When used on the motorized inverted microscopes, these must be compatible with the focus maintenance system (see “Motorized Choices” below). The NA, working distance, and chromatic/planar correction specifications for each magnification are the minimum to meet overall project goals.
Unit #2: 25x Apochromat water dipping objective, NA 1.1, WD 2.0mm, with correction collar for use with or without coverglass. This specialty physiology objective does not need to be compatible with other microscopes, but must support the use of 1300nm light (see “Laser Preferences”). Objectives for other microscopes must be compatible with Unit #2.
Unit #4: 4x Achromat, NA 0.1, WD 30mm; 10x phase contrast achromat, NA 0.25, WD 7mm; 20x phase contrast achromat, NA 0.4, WD 3.9mm. 20 of each objective.
Contrast Preferences:
Unit #2: Widefield Fluorescence, visible scanning confocal, dual-beam multiphoton Unit #4: Phase Contrast, Brightfield Unit #6: Visible scanning confocal, fully compatible and run with the same software as the current Ni-E upright microscope.

1. Coherent Discovery 1300nm dual line laser
1. Autoalignment sytem: translational beam alignment, automated wavelength focal correction optics between 700-1300nm
1. Highest resonant scanning speed 720 fps 512 x 16 Direct Hardware triggering of all devices through confocal/MP controller to eliminate software overhead delays – this is critical for high speed reactions and fast moving proteins
1. Piezo Z minimum range = 450um and must be directly hardware triggered via controller to eliminate software overhead.

1. All diode lasers for enhanced power, stability, and a significantly longer laser lifetime than older gas laser based solutions: such as MultiAr, HeNe, and White light lasers.

2. Minimum power at the fiber tip >=20mW: *this is important since most companies list the power at the laser head which up to 70% can be lost by the time it gets to the fiber tip

Camera, imaging, and analysis software:
All computer workstations must have GPU data processing capability (15x faster than CPU-based processing for analysis algorithms) Software must be same platform for acquisition, processing, analysis, deconvolution, and macro-writing. Ability to switch modes, such as widefield/confocal, point-scanning/resonance, without a software restart.
Unit #4: Color sCMOS camera, full HD (1920x1200) or better, 50fps at full resolution or better, 6µm pixels or smaller. Acquisition software with the same interface as used in all other systems, but only needs capability for capture and simple distance/area measurements.
Unit #5: Must have a CMOS camera supporting a minimum field of view of 25mm, and analysis software that can have custom pre-processing and analysis routines in a saved script to ensure all steps in analysis are repeated the same during each analysis.
Motorized choices:
Unit #3 and #5: Fully motorized nosepiece, light path changing, fluorescence turret, and stage. Nosepiece must include a focus maintenance system that is compatible with all objectives quoted, and capable of functioning with both glass and plastic specimen holders for objectives compatible usable with plastic dishes (long working distance). This system must not expose the specimen to visible light or require imaging to maintain focus, and maintain focus within 1/3 or less the focal depth of the objective. For functionality with thick specimens, the system must support focus across the following ranges above the coverslip or dish bottom:
Air objectives:
4x: +1500 μm 10x: +600 μm 20x: +300 μm 40x: +100 μm
Oil objectives:
40x: +70 μm 60x: +20 μm 100x: +15 μm
The system should be constantly monitoring for focus changes, without software assistance from the PC, and be able to react to any focal change within the specified ranges within 30ms and correct the focus within 700ms from the maximum displacement supported by the objective.
