Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose: NIMH and the trans-NIH Advanced Imaging and Microscopy (AIM) Resource are working together to build a dualview selective plane illumination microscope dedicated to rapidly imaging large samples of chemically cleared brain tissue (CT-DISPIM). This microscope is a modified clone of an existing system at AIM, manufactured by Applied Scientific Instrumentation (ASI). Cloning it, therefore requires purchasing the core hardware from ASI – consisting of the frame, sample stage, principal optics, operating electronics, objective, and cameras.
2. Background Information and Objective: Imaging the distribution of a fluorescent label in brain tissue sections is an essential step for experiments that are being carried out by many basic science investigators within the NIMH intramural program. In order to better preserve the morphology of tissue, it is often necessary to section it coarsely (>1mm) and then optically clear it. The resulting large sections of cleared tissue require many hours or days of imaging on a conventional confocal microscope. Using a light sheet microscope however, reduces the acquisition time to minutes – dramatically accelerating an experimental pipeline. One such light sheet microscope (DISPIM) already exists in AIM and is currently being used to image live or fixed/cleared samples – each of which require a different configuration of the microscope. NIMH is funding the construction of a partial clone of this microscope dedicated for cleared tissue imaging (CT-DISPIM) on which NIMH users would receive priority. The CT-DISPIM is capable of volumetric acquisition from 1-10mm scale tissue samples within minutes with a spatial resolution adequate to trace neurites. The combination of high speed, high volume, and high resolution will be enabling for a variety of projects, including: mapping of injection sites when stereological injections are performed as part of functional opto- or chemo-genetic experiments, mapping projections as part of studies defining the connections of specific neuronal populations, and quantifying changes in gene expression in behavior paradigms.

4. Purchase Description: The CT-DISPIM is purchased as a large set of components that will be assembled on site and will comprise the final microscope. These components that comprise the microscope include:
a) SPIM head set of components;
b) XYZ Stage components;
c) Controller components;
d) Multi-immersion 0.7NA objective;

e) Hamamatsu ORCA Flash 4.0 sCMOS cameras and Camera Link frame grabber cards.
5. Salient characteristics: The CT-DISPIM will have the following key features:
a) Allows single view or dual view light sheet imaging in scanning stage mode;
b) Can image in media with a refractive index in the range 1.33-156.
c) Sub-micron isotropic resolution using 0.7NA multi-immersion objectives;
d) 8 position emission filter wheel;
e) Cameras are capable of 100 frames/second readout with Micromanager controllable synchronization between rolling shutter and light sheet;
f) XY sample stage can travel 120mm x 75mm (xy);
g) All control and acquisition is via ASI DISPIM plugin on Micromanager;
h) Compatible with existing control and analysis software.

8. Period of Performance: All ASI items fully warranted for 5 years, except servomotors in the automated stage which have a 3 year warranty. Equipment is expected to be fully functional for greater than 5 years with minor service, as required.
