RFQ 2017-Q-66463 Attachment 1 Specifications & Instructions to Offerors

Hardware Requirements:

· Cell sorter requires no user optical alignment, laser alignment or laser focus to reduce instrument set-up time and operator interaction in the biosafety cabinet.

· Must have a gel-coupled cuvette that collects 4 times as much excitation light compared to stream-in-air systems thus improving sensitivity in signal detection. (Flow Cytometry Lens System) Sensitivity minimum resolution <118MESF on FITC and 34MESF on PE at 90KhZ and70psi using Sphero rainbow beads.

· Cell sorter BSC maximum dimensions are H 86.5 in, x W 50.2 in, x D 35.6 in.

· Cell sorter must be installed in a NSF-49 certified Biosafety cabinet with ULPA filtration system for live cell sorting. Redundant aerosol management system must function in event of blower motor failure.

· Cell sorter shall incorporate patented octagon shaped collection optics that increase the quality and quantity of the fluorescent signal. Fiber coupled from collection lens.

· Cell sorter to include 405nm,488 nm and 640nm lasers.

· Cell sorter uses digital technology from the preamp forward. The digital electronics uses mathematical calculations in place of analog log amps to provide more linear quantitative measurements.

· Cell sorter must have the ability to threshold on multiple parameters simultaneously. This is a power tool for detecting small particles or samples with large amounts of debris.

· Must have the ability to ratio any signal from any laser.

· Cell Sorter must include a System for Identifying Clusters in Scatter Plots using Smoothed Polygons with Optimal Boundaries

· Cell Sorter must have a cloning unit that can have unique sort regions for different wells with in the same 96 well plate with the ability to chill or heat the 96 well tray.

· Cell sorter must have Index sorting to identify sorted cells on scatter plot.

· Cell sorter must employ automatic sort setup technology that allows the operator to monitor side streams while sorting. This provides for a faster sort set-up providing the operator with a visual display of the sort set-up.

· Cell sorter must have the ability to collect Height, Area and Width parameters simultaneously on all parameters.

· Cell sorter can utilize 6 solid state diode lasers in various wavelengths and outputs lasers and up to 18 colors plus forward and side scatter.

· Cell Sorter must utilize proprietary software that acquires and analyses in bi-exponential display format with FCS 2.or 3.0 export of analyzed data

· Cell sorter must have proven instrument set-up and QC program. This automated Quality Control software package that allows for automation in tracking instrumentation performance. Monitors signal to noise ratio, minimal noise levels for all PMTs and parameters, and allows for feedback on the Q of the laser.

· Cell Sorter must have automated compensation networks between all lasers and all fluoresces parameters.

· Cytometer must be serviceable by in-state factory trained engineers with 48 hour guaranteed on-site repair response time.

· Cell Sorter must self-regulate during sorting. Ability to shut down sample flow, sheath flow, and cover sorted materials in presence of gross shift in sorting parameters. (Flow Sorter With Video-Regulated Droplet Spacing)

· Cell Sorter must have BSC performance conditions so that when operated under normal and stressed conditions (mimicking a clog), <3 Glo Germ™ particles were identified outside the ACDU sort collection chamber. Glo Germ particles, developed by Glo Germ in Moab, Utah, have been shown to provide good visualization of aerosol deposition in normal and mock failure modes by Oberyszyn and Robertson (Cytometry. 43:217-222, 2001).

Software Requirements:

· Software must be compatible with at least Windows 7

· Software must either be approved for use at CDC or a copy of the software must be submitted with the quote for security evaluation at CDC. Purchase of the flow cytometer is dependent on the software passing CDC security evaluation.

· Automated alignment of optics

· Workflow management

· Capable of compensation, quality control, acquisition and analysis

· Data storage and export in at least FCS 3.0 file format

Training Requirements:

· Training for at least 2 CDC personnel

· Of sufficient duration to cover all material, expectation 3 to 5 workdays

· On-site at CDC in the laboratory where the cell sorter is setup

· Includes reagents, supplies and course materials

· Topics covered shall include:

· Overview of instrument components, fluid flow, optics and electronics

· Safety

· Maintenance

· Optics alignment

· Quality Control

· Startup and shutdown of instrument

· Acquisition and analysis

· Gating

· Cell sorting

· Data export

· Service procedures

· Trouble shooting common problems

· Complete test run of sample(s)

Service Agreement Requirements, must include:

· Initial installation

· Shipment costs

· Manufacturer certified replacement parts

· Instrument re-installation following repair

· On-site technical support available in less than 48 hours

· Unlimited service visits during the term of the service agreement on Mondays through Fridays, excluding holidays

· Preventative maintenance

· Telephone technical support

· Software Upgrades

· Option for 1 year agreement

Trade-in Requirements:

CDC would like to trade in a BD FACS Calibur flow cytometer. Any quote should state whether the BD FACS Calibur will be accepted for trade-in and the value of the BD FACS Calibur being credited to the new purchase. All time and expenses for packaging and shipping the BD FACS Calibur will be covered by the provider of the new flow cytometer and not by CDC.

The BD FACS Calibur for trade-in does not include the following:

· Computer system – CPU, monitor, mouse, keyboard

· Performance verification on any component of the instrument or the instrument in total

· May not include other components explicitly stated

Instructions to Offerors

1-The part numbers, where identified, are provided for specification purposes and responses may be either for the specified product or equal item.

2- Contractors are to provide brand name or equal products. If the proposed item does not have the same part number as the one provided, the Contractor must submit enough information to allow the Government to make a determination that the substituted item is of equal specifications. This documentation may include product information pages such as a catalog page. Failure to do so will result in vendor being ineligible for award of those Lines/CLINS. A submission of only the alternate part number and name will not be sufficient.
