Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: The Unit on Neuromodulation and Synaptic Integration (UNSI) seeks to purchase a FVMPE-RS multi-photon microscope system to perform in vivo imaging of neuronal activity with single cell resolution and receptor-based sensors in motivational circuits via the use of GRIN lenses and prisms.

2. Background Information and Objective:

This microscope will allow us to investigate how neuromodulators and in-vivo experiences shape information processing in motivational circuits in-vivo and ex-vivo.

This microscope will allow UNSI lab members to examine single cell activity and dendritic dynamics in head-fixed animals, while simultaneously perturbing circuits.

Moreover, this microscope will be utilized to perform slice electrophysiology experiments while simultaneously monitoring dendritic dynamics using optical approaches with the two-photon microscope. Lastly, this microscope can also be utilized to acquire high-quality images from fixed tissue, which will supplement the capacities afforded by the NIMH Systems Imaging Resource.

3. Generic Name of Product: Complete multi-photon microscope system including set-up and training

4. Purchase Description: 1. Complete multi-photon microscope system 2. FVMPE-RS

(Apollo) Dedicated Resonance Scanning Multiphoton System. 3. Olympus FVMPE-RS (Apollo) Dedicated Resonance Scanning Multiphoton System 4. Model and part numbers as part of quote

5. Salient characteristics for the Laser Scanning multiphoton microscope that shall allow for high-speed and high-resolution 3D Imaging:

1. The scanner head shall have a hybrid dual scanner design that employs both a resonant scanner for high-speed imaging and linear galvanometer scanner for high-resolution imaging.

2. The software must enable users to switch between resonant and galvanometer scanners rapidly.

3. The resonant scanner must allow for an imaging rate of 30 FPS at 512 x 512 pixel resolution. In addition, an optimized flyback control must enable maximum rates of approximately 438 FPS for a clipped scan of 512 x 32 pixels. Non-linear pixel sampling algorithm should provide even illumination and spatially accurate data across full field-of-view.

4. The galvanometer scanner shall define flexible regions of interest, such as points, lines, and shapes, for both imaging and photostimulation. The scanner should also apply a spiral scan pattern to improves photostimulation or bleaching efficiency over a region.

5. Scanner mirrors must have anti-oxidation silver coatings that achieve >90% reflectance from 400 nm to 950 nm, and >95% reflectance from 950 nm to 1300 nm for efficient delivery of excitation light.

6. Scanner lenses must have high transmission coatings that achieve >80% transmission from 400 nm to 950 nm, and >70% transmission from 950 nm to 1300 nm for efficient delivery of excitation light.

7. The scanner head shall be upgradable to a triple scanner design to support independent spatial control of photostimulation via visible or pulsed near-IR lasers.

8. The system must be capable of automated alignment of the excitation laser beam on 4 axes: 2 lateral axes to ensure maximum power throughput, and 2 angular axes to ensure consistent pixel registrations.

9. The system shall automatically compensate for laser pointing direction drift due to wavelength tuning via pre-programmed alignment settings at every 10 nm from 680 nm to 1300 nm.

10. The system should be capable of active alignment using closed-loop feedback based 4 independent position detectors that measure lateral beam position on 2 axes and angular beam direction on 2 axes.

11. The active alignment must be completed in less than 10 s, with a lateral pixel precision of 500 nm at the focal plane when using the 25x objective lens, and 5% precision at the back focal plane of the objective lens.

12. The system shall automatically execute active alignment before z- and t-series acquisitions, and between t-series intervals of 30 s or longer.

13. The system must be capable of software driven manual fine alignment, with an adjustment range of +/- 5 degrees along each angular direction axis, and +/- 36 mm shift along each lateral axis at the back focal plane of the objective lens.

14. The system shall automatically adjust the laser beam diameter using a motorized Kepler beam expander to optimally match the effective back aperture diameter of different objective lenses.

15. The system must have a Deep Focus mode that incrementally reduces the laser beam diameter when imaging deep into highly scattering specimens to produce brighter images, but with reduced resolution.

16. The system must have two high-sensitivity GaAsP PMT detectors that are positioned equidistant to the back focal plane of the objective lens to ensure consistent light collection efficiency between the 2 channels.

17. The GaAsP PMT detectors should utilize fan-less Peltier cooling to minimize signal noise.

18. The detector system must be upgradeable to 2 additional standard-sensitivity or high-sensitivity detectors, for a total of 4 detectors on the epi-collection side.

19. The laser must be a mode locked Ti: sapphire laser with a wavelength tuning range of 690 nm to 1300 nm and a pulse repetition rate of 80 MHz +/- 1 MHz.

20. The laser should have a pulse width of 70 fs or less at 800 nm and a minimum average power of 2.4 W at 800 nm.

21. The laser must have an integrated dispersion compensation system that maintains the short pulse duration at the microscope’s sample plane.

22. The laser has custom output coupling optics that are optimized to match the beam profile to the microscope’s laser delivery optics.

23. The frame is a unique fixed-staged compound microscope frame design devoted to multiphoton microscopy; high degree of motorization, focusing control, fixed and stable.

24. ZDeck quick adjust platform; image multiple areas with speed and precision, maximum stage movement speed = 15mm/sec.

25. MPE specialized spherical aberration correction; critical in deep tissue specimens.

26. Sequencer provides microsecond timing for electrophysiology and optogenetics.

28. The laser must be a dualine laser with tunable excitation from 680–1300 nm and a second fixed line at 1045 nm.

29. Installation fee will include at minimal two 8-hour periods of training.

30. Additional training and support for lifetime of instrument.
