About this file
This document is a justification for other than full and open competition using the brand name exception. It requests 10X Genomics Chromium kits and reagents to conduct single cell RNA sequencing on skin samples obtained from mice. The samples will help characterize the immune response within the tick feeding site by comparing transcriptional profiles of cells from non-pathogen and pathogen-carrying ticks. The justification cites that the 10X Chromium X Controller at the National Institute of Allergy and Infectious Diseases lab can only use proprietary reagents and consumables provided by 10X Genomics as the manufacturer. The related federal contract opportunity is a solicitation from the Department of Health and Human Services National Institutes of Health National Institute of Allergy and Infectious Diseases for four reactions of the Chromium Fixed RNA Kit for the mouse transcriptome.

Justification for Other than Full and Open Competition Brand Name at or Below the Simplified Acquisition Threshold
Project Title: 10X Genomics
I. Identification of the agency and contracting activity
| DHHS, NIH, NIAID, VRC |
| Address of the requesting lab/office: 903 south 4th street Hamilton Montana, 59840 |
II. Description of the project, and supplies or services required to meet the agency’s needs III. Project Description Single cell RNA-sequencing can provide information about the transcriptional profile of every cell within the tick feeding site. The skin samples will be obtained from mice and fixed for fixed RNA profiling. Cluster analysis will be conducted by the bioinformatics team to identify and characterize the cells present within the tick feeding site. We will be able to determine differences in the immune responses by comparing non-pathogen and pathogen-carrying ticks.
A. Description of the supplies or services to be purchased: This request is 10X Gemonics kits.
IV. Identification of the statutory authority permitting other than full and open competition
· 41 USC 253(c)(1) and FAR Part 6.302-1: Only one responsible source
V. Demonstration that the proposed contractor(s) unique qualifications or the nature of the acquisition requires use of the authority cited.
A. Name and address of the proposed contractor(s):
10X Genomics, Inc.
7068 Koll Center Parkway Suite 401 Pleasanton, California 94566 United States
B. Describe why only the requested supply/service will meet the government’s needs:
The Chromium kits and reagents are specific to the 10x Chromium X Controller available for our use; no other kits/reagents are compatible with this instrument, and attempting to run third party reagents would not be supported by the instrument manufacturer.
C. Describe why only the proposed contractor is capable of providing the requested supply/service:
The 10x Chromium X controller system requires proprietary reagents and consumables that can only be provide by the manufacturer, 10x Genomics.
