Justification for Other than Full and Open Competition at or Below the Simplified Acquisition Threshold

Project Title: Chromium X Instrument manf. 10X Genomics Inc.

I. Identification of the agency and contracting activity

DHHS, NIH, NIAID, Center for Human Immunology (CHI)
Address of the requesting lab/office: 10 Center Drive, Bethesda, MD 20892

II. Description of the project, and supplies or services required to meet the agency’s needs A. Project Background The Center for Human Immunology (CHI), part of the NIAID, is involved in the study of characterizing normal and pathogenic human immunome. Studying the protein and gene expression at single cell resolution offers a unique opportunity to inform our understanding of the human immune system. Towards this goal, we have developed several single cell multimodal analysis methods, which combine detection of cell surface proteins, VDJ genes, or chromatin accessibility with transcriptome profiling for tens of thousands of single cells in parallel. They have been applied to multiple studies and more than twenty million cells were analyzed in the last years. The 10X Genomics Chromium X has the platform for the HT assay.

B. Project Description The 10X Genomics Chromium system is high throughput and the reagents and protocol can cover much more cells at one time, increasing processing time and meets the demands of our increased workload.

III. Identification of the statutory authority permitting other than full and open competition ☒ 41 UCS 1901(a)(1) and FAR Part 13.106-1: Only one responsible source

IV. Demonstration that the proposed contractor(s) unique qualifications or the nature of the acquisition requires use of the authority cited.

A. Name and address of the proposed contractor(s) :

10x Genomics, Inc.

6230 Stoneridge Mall Road Pleasanton, California, 94588-3260 United States B. Describe why only the requested supply/service will meet the government’s needs The 10X Genomics Chromium system is high throughput and the reagents and protocol can cover much more cells at one time, increasing processing time and meets the demands of our increased workload.

The Chromium X has the following required features:

· Multiple sensors, very accurate pressure control, and temperature controlled tray.

· Firmware upgrades, network connectivity and syncing to the 10x Genomics Cloud, to log run information and experimental setups.

· Similar performance as assessed by multiplet rates, library complexity, read mapping rates, and cell clustering.

· Ability to run an assay with low throughput (LT), standard, or high throughput (HT).

· Low throughput: 100–1,000 cells per channel

· Standard assay: 500-10,000 cells per channel 500-30,000 cells per channel with CellPlex

· High throughput (HT): 2,000–20,000 cells per channel 2,000–60,000 cells per channel with CellPlex

· Works with the standard assays we have been using in last few years.
