Attachment I – Specifications
1 | P a g e
I. BACKGROUND
The Biomedical Microtechnologies Group within the Microsystems and Nanotechnologies Division of the Physical Measurement Laboratory, in coordination with the NIST-on-a-chip program, conducts research on miniaturized, “lab on a chip” (LOC) technologies with embedded sensors. The Biomedical Microtechnologies Group builds custom microfluidic cytometry measurement tools and needs to purchase a commercial fluorescence activated cell sorter (FACS) that can refine cellular populations through sorting and serve as a tool for establishing and improving uncertainty quantification for the field of flow cytometry. More specifically, NIST intends to use the commercial system to measure and sort populations of fluorescent beads and cells that will then be analyzed in our custom cytometer. The sorter will also be used for more traditional measurements and sorting of cells for research projects related to development of other technologies such as body-on-a-chip systems. Samples will include lymphocytes, liver and lung cells, breast cancer lines, and stem cells. Given the breadth of NIST’s research portfolio and lack of cytometry technician, NIST requires a system that is easy to use and maintain and includes automated and straightforward startup and flow optimization and intuitive measurement and characterization tools.
NIST’s target measurements require a system with 3 lasers (405 nm, 488 nm, and 642 nm) and multiple fluorescence emission channels to discriminate a wide variety of commercially available fluorophores that are excited by these wavelengths. For example: Alexa488, DAPI, Cy5, Cy7, PE, APC, Brilliant Violet, and many other single and tandem dyes. In order to reduce the effects of spectral overlap, NIST requires a system that has spatially separated lasers and well separated emission bands. NIST’s needs also require a biosafety cabinet enclosure for safe containment of biohazards during operation.
II. MINIMUM TECHNICAL SPECIFICATIONS
The system shall meet the minimum requirements identified in the Specifications Document. All items must be new. Used or remanufactured equipment will not be considered for award.
Experimental, or prototype will not be considered. The use of “gray market” components not authorized for sale in the U.S. by the proposer is not acceptable. All line items shall be shipped in the original manufacturer’s packaging and include all original documentation and software, when applicable.
The required sorting flow cytometer shall be capable of meeting the following requirements:
1) Optical configuration, detection, and fluidics
a. Lasers: 3 lasers to excite fluorescent dyes normally excited by 405nm, 488 nm, and 640 nm wavelengths
• ≥ 20 mW output power
• Lasers must be spatially separated in order to minimize spectral crossover
b. Detectors: all detectors must have capability of extracting signal width, height, and integrated area.
• All must have at least 18-bit resolution.
• Gain control to optimize signal to noise ratio on each detector.
c. Sensitivity: Must have high fluorescence sensitivity; demonstrated Mean Equivalent Standard Fluorophores (MESF) of <125 (FITC)
d. Scatter collection: Forward and side scatter collection channels
e. Fluorescence Detection: At least nine (9) fluorescence detection channels to cover dyes similar to those listed below (approx. bandpass filter center/width wavelength in nm):
i. 405 nm laser (3): DAPI (488/45), BD Horizon V500 (528/45), Brilliant Violet
786 (752nm longpass dichroic)

g. Monitor
Warranty The Contractor shall provide, at a minimum, a one (1) year warranty for the equipment. The warranty shall cover all parts. The Contractor shall provide OEM certified service technicians for the warranty period. The warranty shall commence upon successful completion of delivery, installation, training, and the Government’s acceptance.
III. DELIVERY
Delivery terms shall be F.O.B. Destination. Scheduling of delivery shall take place at a time mutually agreed upon by the Government and the Contractor. The delivery (inclusive of installation and training services) shall be completed no later than 130-days after award.

IV. INSTALLATION
The Contractor shall install the system at the NIST Gaithersburg, MD campus. Installation, at a minimum, shall include uncrating/unpackaging of all items, rigging, set-up and hook-up of the equipment, demonstration of all specifications, and removal of trash.
V. TRAINING
The Contractor shall schedule and facilitate one (1) training session on-site at NIST Gaithersburg, MD.
The training shall provide a thorough demonstration of all system functions, system operation, and basic troubleshooting. The training may be completed on-site at NIST immediately following installation and demonstration of specifications. Training shall be coordinated with the NIST Technical Point of Contact (TPOC), to be identified at the time of award, to ensure maximum availability of NIST personnel.
VI. INSPECTION & ACCEPTANCE
The Government will complete inspection and acceptance in accordance with Federal Acquisition Regulation (FAR) 52.212-4(a). The Government anticipates that inspection and acceptance will be completed within ten (10) business days upon completion of delivery, installation, and training.
A visual inspection of the system will be performed by the NIST TPOC to identify surface defects or any form of indication that the system was damaged during transport to NIST. The Government shall have sole discretion to require repair or replacement of damaged and/or nonconforming supplies at no cost to the Government. The Government at any time prior to acceptance shall reject the system due to defects and/or nonconformance.
