Specifications Automated Single Cell Dispenser
I. BACKGROUND:
BBD is focused on developing methods that will assure reliability and confidence in the measurements that underpin research and applications in genomic measurements, biomaterials, protein therapeutics and cell therapies, cancer diagnostics, nanoparticle safety, and microbial identification and quantification. We carry out this work through fundamental research in measurement science and the development of reference materials, reference data, and protocols.
II. SPECIFICATIONS:
The contractor shall deliver an automated single cell dispenser with the following specifications:
· Must be use as an event detection mechanism to directly count 1 to 100 cells/well in a SBS format 96-well and 384-well plates.
· Must contain at least one event detection mechanism that can detect unlabeled/unperturbed cells.
· Must be able to demonstrate >75% viability of dispensed cells with a CHO or an NIH 3T3 cell line.
· Must be able to sort cells that are between 5 and 35um in diameter.
· Must use low pressure fluidics for sorting (less than 4 psi)
· Must fit into a sterile BSL2 hood with a minimum 120 cm x 46 cm working surface
· Must include software and computer system that allows video or other documented evidence that a cell has been dispensed into a well.
· Must include software that stores cell dispensing documentation evidence on a computer for post-dispensing analysis of the cells that were dispensed into a well.
· Must include software that provides option for software to generate
· Scatterplots/histograms for cell level data
· Gaiting on cell size and cell shape and other cell properties
· Must save documentation data in a format that can be read by spreadsheet software (i.e. Microsoft Excel) or third-party image processing software (i.e. ImageJ)
· Must accommodate disposable sterile cartridges for the cell suspension making rapid clean up and instrument reuse possible.
· Must include 10 disposable sterile cartridges that would allow 10 cell dispensing experiments to be performed with 96-well plates.
· Must be able to accommodate 70% EtOH or equivalent isopropanol wash steps to sterilize tubing and pump parts for maintenance.
· Must provide a computer and monitor that meets or exceeds requirements for instrument operation
· Must include at minimum, a two-year warranty
· Shall provide installation and training at NIST facility in Gaithersburg, MD
III. DELIVERY & INSTALLATION:
Delivery of the system shall occur 30 days after award; installation and training shall be completed within 45 days after award.
IV. TRAINING:
The contractor shall provide one (1) training session for NIST personnel. The training shall provide a thorough demonstration of all equipment functions, operation, and basic user-level troubleshooting features. The training shall be completed on-site NIST within 45 days of award.
V. WARRANTY:
The contractor shall provide at a minimum, a two-year warranty for the equipment. The warranty shall cover all parts, labor, and shipping. The warranty shall commence upon successful completion of delivery, installation, and performance of the specifications.
VI. GENERAL INFORMATION:
Safety: The Contractor employee shall be responsible for knowing and complying with all installation safety prevention regulations. Such regulations include, but are not limited to, general safety, fire prevention, and waste disposal.
Security: NIST is a restricted campus. An identification badge is required for access for entry into buildings and also is shown to the armed Security Police when entering the campus.
Regular Business Hours: Regular business hours are Monday through Friday, 8:00 am to 5:00 pm Eastern Time.
