Statement of Work (SOW) Project: Bordetella Genome Sequencing

GENERAL INFORMATION

1.0 Scope of Work: Hi-C analysis of DNA from a bison bull for the purpose of enhancing the de novo assembly of the bison genome (2.82 Gb genome size).

2.0 Background: This project will use data from Hi-C, a technique based on proximity ligation in which DNA sequence segments that are physically close to one another in the nucleus are cleaved by a restriction enzyme and preferentially ligated together in situ before DNA is isolated from cells. This allows identification of DNA segments (contigs) that are in close association and facilitates closures of gaps and misalignment of on contigs. Use of this technique allows closure of gaps in a genomic sequence that cannot be closed by other methods. .

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

1) Provider must perform Hi-C analsysi including ligation in situ, DNA isolation, library preparation, sequencing, and data delivery for DNA samples.

2) Provider must assess/measure the samples provided by the client to ensure that the samples meet quality thresholds. In the event the sample is not of sufficient quality, the provider will notify the client to obtain a new sample..

3) Provider will do a Hi-C libarary preparation, obtain low coverage sequencing on the library for quality control purposes (1M paired end illumina reads), conduct deep sequencing of the Hi-C library, do a prroximo assembly of the genome, and conduct a genome haplotyping analysis of the assembled genome.

4) The client will provide the provider access to the current bison genome assembly.

5) Provider must electronically transfer Hi-C data to the client and/or the bioinformaticist working with the client.

4.0 Government Furnished: A frozen blood sample will be furnished to the provider

5.0 Deliverables / Schedule: Provider will provide Hi-C analysis via electronic data transfer or via external hard drive.

6.0 Travel: None required

7.0 Contractor’s Key Personnel

8.0 Security Requirements: None required

9.0 Data Rights: Data rights will be transferred to the researcher upon completion of the sequencing project.
