STATEMENT OF WORK FOR nCOUNTER COMPATIBLE CUSTOM NUCLEIC ACID DETECTION REAGENTS
I: GENERAL INFORMATION
A. Introduction: The U.S. Geological Survey, Leetown Science Center, National Fish Health Research
Laboratory requires bioinformatic design and synthesis of a custom set of hybridization probes for the quantification of RNA molecules in tissue lysate using the SPRINT nCounter Analysis system. The scale of the project is 192 reactions (samples) and 40 unique genes targets.
B. Background: The National Fish Health Research Laboratory and the Fish Health Branch (FHB) are part of the U.S. Geological Survey’s Leetown Science Center (LSC), located at 11649 Leetown Road, Kearneysville, WV 25430. The NFHRL conducts health and disease research on fish and wildlife for
Department of the Interior bureaus and other Federal and State agencies. The research includes study of both infectious and non-infectious diseases of fishes and other aquatic organisms and includes characterization of infection, molecular pathogenesis, proteins, hormones and disease processes in aquatic organisms as these factors relate to the host, pathogen, and environment.
C. Definitions: Multiplex – assessment of multiple gene targets in a single sample
D. Scope: The purpose is to design gene specific probes based on in-house sequence data and digitally count specific RNA molecules in tissue lysate using the SPRINT nCounter platform that will be used to evaluate the expression of immune-relevant genes in smallmouth bass leukocytes.
Must be able to design a multiplex nucleic acid detection probe set using in-house (user supplied) nucleotide sequence libraries.
Probe set must be capable of uniquely discriminating 40 genes in a single sample
Data output from the detection reaction must be compatible with digital detection
Probes and reaction buffers must be compatible with the SPRINT nCounter
The multiplex reaction must be compatible with tissue lysate
Detection probes should not require enzyme-based amplification
II. WORK REQUIREMENTS
A. Technical Requirements:
Custom detection probes must be compatible with the SPRINT nCounter
Contractor shall ship custom probes and necessary reaction buffers
Contractor will provide a minimum of one-year warranty on the stability and functionality of the reagents stored at proper conditions.
Contractor will provide all warranties and protocols for the detection reagents
B. Criteria for Acceptance of Product or Service: N/A
C. Deliverables: The contractor shall provide deliverables in accordance with the Reporting
Requirements Checklist.
III. SUPPORTING INFORMATION:
A. Place of Performance: Custom nucleic acid detection reagents will be delivered to the U.S. Geological
Survey, Leetown Science Center, Fish Health Lab located at 11649 Leetown Road, Kearneysville, WV
25430.
B. Period of Performance: The contractor shall deliver the detection reagents within 60 days of receiving target sequences from scientists at the Leetown Science Center.
C. Government Furnished Property: N/A
