Duke University has been awarded a $4,537,430 Project Grant from the National Institute on Aging, part of the Department of Health and Human Services, to support research under the Aging Research program (CFDA 93.866).
The grant will fund a study from September 2022 to August 2025 to examine the role of cellular senescence on postoperative neuroinflammation. Specifically, the researchers will use a mouse model of orthopedic fracture to determine how senescent cell burden exacerbates systemic inflammation, blood-brain barrier dysfunction, and neuroinflammation leading to conditions like delirium. They will also test the efficacy of senolytic drugs to selectively eliminate senescent cells in preventing neuroinflammation. Finally, the study aims to better understand the interactions between blood-borne factors and the blood-brain barrier using microphysiological systems. The research is intended to provide new insights into the effects of peripheral surgical trauma on the brain and identify potential pharmacological targets to improve outcomes for elderly patients undergoing surgery.
