This $535,743 Project Grant award from the National Institute of Neurological Disorders and Stroke (NINDS), under the Extramural Research Programs in the Neurosciences and Neurological Disorders (CFDA 93.853) program, supports research at the University of Wisconsin-Madison to investigate the mechanisms controlling cargo transport and localization in developing sensory neurons. The key objectives are to: Determine the mechanisms by which the kinesin light chain 4 (KLC4) protein regulates axon branching and compartmentalization, and the organelle cargos it transports. Examine the functions of KLC2 in axon development and cargo transport, and the underlying mechanisms of the human spastic paraplegia (SPOAN) syndrome. Identify the protein domains and adaptors that confer functional specificity to different KLC proteins during neuronal development and axon growth. This research aims to elucidate the molecular signals regulating neuronal compartmentalization, cargo selection/transport, and axon growth/branching, with potential to inform treatments for developmental and neurodegenerative disorders. The project period runs from August 1, 2023 to July 31, 2024.
