The University of Texas Health Science Center at San Antonio (UTHSCSA) was awarded a $408,179 Project Grant from the National Institute of General Medical Sciences (NIGMS) Biomedical Research and Research Training program (CFDA 93.859) to conduct research aimed at defining the functional interactome of DNA damage response (DDR) proteins and mechanistically delineating post-translational modification circuitries to better understand DNA repair mechanisms.

Specifically, the project will investigate the role of the ARAF kinase in regulating DDR, examine DNA repair defects associated with ARAF depletion or inhibition, and analyze the spectrum and kinetics of the phosphatase activity of spinophilin (SPN)-PP1 complexes within DDR. Additionally, the researchers will investigate whether the histone demethylase RSBN1L is recruited by BRCA1-BARD1 complexes to DNA double-strand break sites to regulate the choice between homologous recombination and non-homologous end joining repair pathways. This 5-year research program is expected to provide novel insights into the dynamic protein-protein interactions and post-translational modifications that govern the coordinated DDR.
