This Project Grant award of $399,829.00, provided by the National Cancer Institute (CFDA 93.394 - Cancer Detection and Diagnosis Research), aims to develop novel glioblastoma (GBM) models to investigate the impact of sex chromosomes and sex hormones on GBM phenotype, including proliferation and cellular metabolism. The award will fund two key objectives: 1) Using the Four Core Genotypes mouse model, the researchers will define the contributions of sex chromosomes and in utero sex hormone exposure to astrocyte metabolism before and after GBM transformation; and 2) Developing human induced pluripotent stem cell models to evaluate the effects of sex chromosomes on cellular differentiation and mesenchymal GBM transformation. The research will leverage advanced diffusion MRI imaging to non-invasively assess changes in the tumor microenvironment. This innovative work is expected to provide critical insights into the drivers of sex disparities observed in GBM incidence and mortality, with the goal of informing future research and therapeutic strategies.
