This Project Grant award, funded by the National Cancer Institute (CFDA 93.395 - Cancer Treatment Research), aims to develop a phage-based gene delivery system to target and modulate the tumor microenvironment in triple-negative breast cancer (TNBC). The 2-year, $403,686 award will support research at Rutgers, The State University of New Jersey, to explore the use of a targeted hybrid vector, AAVP, to deliver the human tumor necrosis factor (TNF) gene to TNBC tumor cells and associated vasculature, as well as tumor-associated macrophages. The goal is to repolarize the immunosuppressive M2-like tumor-associated macrophages to a pro-inflammatory M1-like phenotype, reversing immune subversion and potentially enhancing the efficacy of existing TNBC treatments. The proposed approach combines the advantageous biological attributes of eukaryotic and prokaryotic viruses to facilitate targeted systemic gene therapy for this aggressive breast cancer subtype.
