The National Heart, Lung, and Blood Institute (NHLBI) awarded a $805,845 Project Grant under the Cardiovascular Diseases Research program (CFDA 93.837) to the Proteogenomics Research Institute For Systems Medicine in La Jolla, CA. The goal of this 4-year project is to develop and test a novel "Lung Precision Drug Delivery System" (LPDDS) that can actively and specifically transport myofibroblast-targeted antifibrotic therapeutics directly into lung tissue. The LPDDS leverages caveolae-mediated transcytosis to enhance delivery and penetration of therapeutics like the bispecific antibody construct MAPP:THY-1 and the immunoconjugate MAPP:FAP-nintedanib into the lung interstitium. This approach aims to more effectively target and reverse pulmonary fibrosis by concentrating antifibrotic activities at the site of disease. The project will optimize the LPDDS, assess its lung tissue delivery and therapeutic efficacy in rodent models, and lay the groundwork for caveolae-targeted therapies that could shift the paradigm for pulmonary disease treatment from passive to active drug delivery.
