This award supports the development of drug-loaded degradable biopolymeric scaffolds designed to augment preoperative liver growth in cancer patients undergoing surgical resection. The research addresses critical clinical challenges associated with insufficient future liver remnant (FLR) volume following hepatic tumor resection, which significantly increases the risk of post-hepatectomy liver failure and associated morbidity and mortality. The project delivers three primary research outputs: (1) development of a rat tumor model with cirrhosis background to enable translational research; (2) fabrication of resorbable polymeric scaffolds optimized for sustained delivery of obeticholic acid (OCA), a bile acid analogue that promotes hepatocyte proliferation and reduces inflammation and fibrosis; and (3) implementation of positron emission tomography (PET) imaging using the novel radiotracer [18F]FSPG to non-invasively monitor postintervention FLR growth and tumor progression. Research findings will be correlated with histologic and transcriptomic liver and tumor profiles to elucidate mechanisms of action and establish clinical translatability of the delivery system and imaging modality.Federal Grant Award Summary
