This Project Grant award from the National Cancer Institute's Cancer Biology Research program (CFDA 93.396) provides $468,139 to Tulane University to investigate the immunosuppressive function of the long non-coding RNA CRNDE in hepatocellular carcinoma (HCC).
The key objectives are to: 1) elucidate the molecular mechanisms by which HCC-derived CRNDE inhibits the anti-tumor immune response of tumor-infiltrating lymphocytes (TILs) and promotes tumor growth; and 2) determine the in vivo anti-HCC efficacy of CRNDE inhibition in mouse models. The research will employ complementary mouse models, ELISPOTs, flow cytometry, single-cell RNA sequencing, and other techniques to analyze TILs and delineate the CRNDE-CYPB interaction and its effects on NFATC signaling.
The project aims to uncover a previously unrecognized mechanism of CYPB regulation in HCC immune evasion, and develop a new CRNDE-targeted approach for HCC immunotherapy. The award period is from March 15, 2025 to February 28, 2030.
