This $3.276 million project grant from the Department of Health and Human Services Assistant Secretary for Financial Resources will support research at St. Jude Children's Research Hospital Inc. from September 2022 through August 2025 under the Biomedical Research and Research Training program (CFDA 93.859).

The research aims to advance understanding of dynamic spatial organization and subcellular architecture as it relates to metabolism through multidisciplinary approaches. Investigators will apply whole-cell focused ion beam scanning electron microscopy and deep learning techniques to systematically examine and reconstruct subcellular organization during metabolic reprogramming events at high resolution. This will enable mathematical modeling of architectural features associated with specific metabolic conditions and diseases. Additionally, the project will develop a platform using reversible split fluorescent proteins to visualize and quantitatively monitor organelle contact sites implicated in directing inter-organelle material flow and thereby impacting metabolic outcomes under different conditions. Findings are expected to significantly transform knowledge of architecture-function relationships in cell biology and generate new research directions.
